Abstract
Objective To establish a high performance liquid chromatography(HPLC) method for the determination of mycophenolic acid(MPA) in children's plasma in order to monitor the plasma drug concentration of mycophenolate mofetil(MMF). Methods The determination by HPLC was carried out using Waters Spherisorb C18 column(ODS 4.6 mm×150 mm, 5 μm), with mobile phase of acetonitrile: water: triethylamine(40:60:0.3)(Phosphoric acid adjust pH to 3.5) at a flow rate of 1.0 mL/min. Column temperature 25℃. The detection wavelength was 252 nm. Results Excellent linear correlation was established within the range of 0.5 ~ 60 mg/L(r = 0.996). The absolute recovery of high, middle and low concentrations were 91.5 %, 93.7 %and 96.9 %, respectively, and relative recoveries were 103.3 %, 95.6 %and 104.7 %, respectively. The RSDs difference between intra- day and inter- day were less than 5 %. 20 samples of children patient has been determined by developed HPLC, and the sample concentration were 0.76 ~ 2.67 mg/L. Conclusion This method is convenient and accurate for monitoring drugs concentration in blood samples of children patients.
