Interaction of intercellular adhesion molecule-1 gene polymorphisms and other exposure factors on coronary heart disease

Abstract
Objective To investigate the association between intercellular adhesion molecule-1 (ICAM-1) gene polymorphisms and coronary heart disease (CHD).Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and PCR sequence-specific primers (PCR-SSP) were used for the detection of ICAM-1 genotypes in 211 patients with CHD and 206 healthy controls. Multiple Logistic regression analysis was made to explore the risk factors for CHD.Results The frequencies of KK genotype and the K allele were significantly higher in CHD group than that of controls (55.5% vs 44.7%, P0.05; 74.9% vs 67.7%, P0.05). The relative risk suffered from CHD of KK genotype was 1.542 times of the EE and KE genotype (OR=1.542,95%CI:1.048~2.269). Logistic regression analysis showed that there were correlation between CHD and hypertension, diabetes mellitus, genotype KK, abusing cigarette, TG WBC counting.It also showed a significant gene-environment interaction (P0.05). The OR value were: for KK genotype-hypertension: 21.02 (95%CI:4.72~93.59, P0.01), for KK-DM: 26.89 (95%CI:1.93~373.83, P0.01),for KK-smoking: 5.45 (95%CI:1.74~17.03, P0.01). However, we didn’t detect the G241R polymorphism in our study.Conclusions The coding single nucleotide polymorphism (cSNP) at codon 469 in the exon 6 of ICAM-1 gene is associated with CHD and the allele K may be a risk factor for CHD in Chinese. The carriers of KK genotype would have higher risk of suffering from CHD if they also suffer from hypertension or diabetes or they abuse cigarette. There is no G241R polymorphism in Chinese.
