Effects of Macleaya cordata Alkaloids on Intestinal Porcine Epithelial Cells Proliferation

Abstract

To study the effects of sanguinarine,chelerythrine,protopine and allocryptopine from Macleaya cordata on intestinal porcine epithelial cells(IPEC-1) proliferation at different concentrations and different periods.IPEC-1 cells were pre-incubated for 24 hours,and then exposed to different concentrations(4,2,1,0.5,0.25,0.1,0.05,0.025,0.012 5 and 0.006 25 μg/mL of sanguinarine or chelerythrine,and 100,50,25,12.5,6.25,3.125 and 1.562 5 μg/mL of protopine or allocryptopine;each treatment had 8 replicates)for additional 12 or 24 hours,respectively,cell viability was measured by MTT assay.The results showed as follows:after the IPEC-1 cells were treated by 0.25 to 4 μg/mL of sanguinarine,0.25 to 4 μg/mL of chelerythrine,100 μg/mL of protopine or 100 μg/mL of allocryptopine for 12 hours,the proliferation of cells was significantly inhibited(P0.05);whereas 0.006 25 to 0.1 μg/mL of sanguinarine,0.006 25 to 0.1 μg/mL of chelerythrine,3.125 to 25 μg/mL of protopine or 1.562 5 to 25 μg/mL of allocryptopine significantly promoted IPEC-1 proliferation(P0.05),however,with Macleaya cordata alkaloids concentrations decreasing,the effects had slowly diminished;when extended the treatment time to 24 hours,only 0.1 μg/mL of chelerythrine,25 μg/mL of allocryptopine and 25 μg/mL of protopine showed a significant enhance effect on the IPEC-1 proliferation(P0.05),there were no significant differences between the other experimental groups and control group(P0.05).The results indicate that the Macleaya cordata alkaloids can promote the proliferation of IPEC-1 under certain concentrations;it speculates that Macleaya cordata alkaloids may improve the intestinal digestive,absorptive,immune and barrier function through this way,and then improve the animal growth performance.
