Abstract
Objective To study the histopathological changes and ultrastructural changes in the substantia nigra of Parkinson's disease rats and to investigate the role of these changes in the pathogenesis of PD.Methods PD rat model was created by stereotaxic injection of 6-OHDA into the left substantia nigra pars compacta(SNC) and ventral tegmental area(VTA).Histopathological changes and ultrastructural changes in the substantia nigra were examined after the injection of 6-OHDA.Results The number of tyrosine hydroxylase immunopositive neurons in the left(the injured side) SNC of PD rats significantly decreased in experimental group compared to the right side throughout the test time.The number of tyrosine hydroxylase immunopositive neurons in the left SNC 2 or 4 weeks after injection significantly decreased when compared to 1 week after injection in PD group.The nigral neurons of rats in PD group showed karyopyknosis,mitochondria swollen,crista lost,dilatated endoplasmic reticulum,obvious degranulation,blurring membranes of pre-synapsis and post-synapsis and disappeared synaptic space.No changes of morphology were found in the control rats.Conclusion Tyrosine hydroxylase immunopositive neurons significantly reduce in the injured SNC of PD rats.There are significant ultrastructural changes in SNC,which may contribute to the death and loss of neurons in PD.
