Abstract

[Objective] The aim was to study the optimum culture conditions of tomato callus induction and cell suspension culture. [Method] Taking tomato cotyledon as explants, the effects of different concentration combinations with 2,4-D and 6-BA on callus formation and differentiation of adventitious buds were studied. [Result] MS medium added to different concentration 2,4-D and 6-BA had different effects on callus formation. Thereinto, hormones played a major role. The best medium of callus induction was MS + 2,4-D 0. 05 mg/ L + 6-BA 0. 01 mg/ L, which had good effects on callus formation and differentiation of adventitious buds. The different hormones had some effects on cell suspension culture, and the best combination of concentration was 2,4-D 0.2 mg/ L+NAA 0.2 mg/ L+ZT 0.05 mg/ L. [Conclusion] The study confirmed the optimum culture conditions of tomato callus induction and cell suspension culture, and provided basis for tomato tissue culture.
