Abstract
Objective: To study the expressions of GRP78 and GADD153 and their mutual correlations in palatal mucosal carcinogenesis of Wistar Rats.Methods: 70 Wistar rats were equally divided into 10 groups,with one group as the control.A chemical carcinogen,4NQO,was used to induce oral cancer in the other nine groups.The expression levels of Grp78 and GADD153 were detected by the immunohistochemistry S-P method,and mean optical density(OD) of positive cells were measured by image analysis.Spearman correlation analysis was then performed to find out the relations between the two kinds of measuring results.Results: The OD of Grp78 positive cells increased gradually as the histopathological grading increased.Difference was found between normal mucosa,dysplasia and oral carcinoma groups(P0.01).The OD of GADD153 positive cells in mild,moderate,severe hyperplasia of the mucosa and oral carcinoma decreased gradually(P0.01).Negative relations was found between the two ODs of Grp78 and GADD153 after Spearman analysis(P﹤0.05).Conclusion: Grp78 and GADD153 may play some roles in oral mucosal carcinogenesis.The tests of both proteins can help the early diagnosis and gene treatment on oral carcinoma.
