Abstract

Objective To observe the effects of hyperglycemia on the expression of TGF-β1,CTGF,and to explore hyperglycemia and correlation between hepatic fibrosis. Methods 40 SD rats were randomly divided into four groups according to body weight,n = 10,the control group( C group),hyperglycemia group( H group),liver fibrosis group( M),hyperglycemia + liver fibrosis group( H + M group). The liver fibrosis was made with carbon tetrachloride liver fibrosis model,with the emergence of streptozotocin-induced hyperglycemia. All the animals were killed after eight weeks. The TGF-β1 and CTGF levels were detected by ELISA. The TGF-β1 and CTGF mRNA expression were detected by real-time quantitative RT-PCR. Results TGF-β1,CTGF levels and mRNA expression in group C and H group did not change significantly( P 0.05). Serum levels of these parameters of M and H + M group were significantly higher than that in group C( P 0.05). While the serum these indicators of H + M group increased more significantly( P 0. 05). TGF-β1 and CTGF has a significant positive correlation( r =0. 812,P 0. 05). Conclusion High blood sugar can increase serum liver fibrosis TGF-β1 and CTGF expression and content. The TGF-β1and CTGF content and degree of liver fibrosis has a positive correlation between the evaluation of liver fibrosis. These indexes can be used as an important indicator.
