Abstract

Objective To investigate the relationship between the cultured dermal papilla cells in aggregative growth pattern and syntheses of their extracellular matrix.Methods Histochemical technique with Alcian blue staining and PAS reaction was employed to show the intensitis of extracellular matrix in the human dermal papilla cultured in DMEM medium with different pH value Results The intensities of staining in dermal papillae cultured in DMEM medium at pH 6.8 showed a significant red with PAS.The intensities of staining in dermal papillae cultured in DMEM medium at the pH value of 7.2and 7.6 showed either red or blue.Aggregative growth pattern was observed only in dermal papilla cells cultured in medium at the pH value of 7.6 and7.2.The intensities of staining in dermal papillae cultured in DMEM medium at the pH value of 8.0 showed only red.Aggregative growth pattern was not observed in dermal papilla cells cultured in medium at the pH value of 8.0.Conclusion The study suggests that the best pH value of culturing environment is 7.2-7.6.The pH value of culturing environment affects the syntheses of the extracecllular matrix in dermal papilla cells The syntheses of acid mucopolysaccharide has a very close relation with aggregative growth pattern of dermal papilla cells.
