Abstract
Objective To prepare film-controlled sustained release pellets of glipizide by studying itsin-vitro release profiles. Methods Incorporation of the drug into pellets was performed by centrifugal granulation technology, using the powder layering theory, and then the pellets were coated with EUDRAGIT by fluid-bed coating method to prepare a 12 h sustained-release preparation. Its in-vitro release was compared with that of the matrix sustained release preparations available in the market. Results The in-vitro release profile of glipizide film-controlled sustained-release pellets fitted the first-order model. Burst drug release effect was obvious for matrix sustained release pellets. Conclusions The glipizide film-controlled sustained-release pellets were successfully prepared and the in-vitro drug release conformed to the requirements of the sustained-release formulation.
