Experimental study of nerve allograft through the application of slow-releasing film with FK506/NGF/RGD composites

Abstract

Objective:To investigate the effect of FK506/NGF/RGD slow-releasing film on nerve allograft.Methods:Forty-eight male SD rats were used in this study,involving eight rats as the donors.Bilateral sciatic nerves were taken out from the rats and cryopreserved in-196℃ liquid nitrogen for three weeks.Forty rats were randomly divided into four groups.Group A:cold preservation of nerve allograft;Group B:cold preservation of nerve allograft with FK506/RGD slow-releasing film;Group C:autograft;GroupD:cold preservation of nerve allograft with FK506/NGF/RGD slow-releasing film.Twelve weeks after operation,the results were evaluated by morphology,electrophysiology,weight of gastrocnemius,histology,ultrastructure etc.Results:The acute reject reaction was not found after operation.The results of electrophysiology and histology showed that the regeneration nerve in group D was obviously better than that of other groups.Conclusions:The application of FK506/NGF/RGD slow-releasing film to nerve allograft can accelerate nerve regeneration effectively.
