Abstract
The polysaccharide isolated from the spores of Ganoderma lucidum was purified and characterized. The crude polysaccharide was obtained from the hot-water extract of the spores of Ganoderma lucidum and the polysaccharide SGL-Ⅱ-2 was isolated by Sepharose CL-6B column chromatography. The HPLC analysis indicated that the SGL-Ⅱ-2 was a homogeneous polysaccharide. Its molecular weight was estimated to be 5.37×10 4.By means of partial hydrolysis with acid, periodate oxidation,Smith degradation,methylation,IR,GC,GC-MS and 13 C NMR,the structure of SGL-Ⅱ-2 was identified. It is a lowly branched molecule with basic structure composed of (1→3)-linked-glucose and (1→6)-linked-glucose residues in the main chain. Some of (1→6)-linked-glucose residues are substituted at 3-O or 4-O.The side chain is composed of (1→4)-linked-galactose.The nonreduced end is glucose.
