Abstract
Objective:To observe the inhibitory effect of sulindac on the growth of gastric cancer cells BGC-823 and its antineoplastic mechanisms.Methods:Human gastric cancer cells BGC-823 were cultured with various concentrations of sulindac for different time.The proliferation of BGC-823 cells was measured by MTT colorimetrie assay;and the cell cycle distribution was examined by flow cytometry.Cell apoptosis was determind by TUNEL.The expressions of ki-67,survivin and COX-2 in cells were detected by immunohistochemical staining.Results:Sulindac inhibited proliferation of BGC-823 cells,increased the proportion of cells in the G_0/G_1 phase,decreased the proportion of cells in the S phase,induced apoptosis of the BGC-823 cell line and decreased the expressions of ki-67,survivin and COX-2 in cells.All the effects were in a time-dependent and dose-dependent manner(P0.05).Conclusions:Sulindac inhibits the growth of gastric cancer cells BGC-823 and the antitumor mechanism may be related to changing cell cycle distribution,inducing cell apoptosis and inhibiting the expression of COX-2,survivin and ki-67.
