Effects of gastric bypass on GLP-1 secretion and blood glucose in type 2 diabetic rats

Abstract

Objective To investigate the mechanism of gastric bypass surgery (gastric bypass) for the treatment of type 2 diabetic (non-insulin dependent diabetes mellitus, NIDDM) rats. Methods Eight-week-age Wistar rats were randomly divided into four groups: type 2 diabetes-operation group (DO group) and type 2 diabetes-control group (DC group), normal-operation group (NO group) and Normal-control group (NC group). The blood glucose and glucagon-like peptide-1 (GLP-1) were recorded and measured respectively at fasting and post oral glucose 30 min condition. Results The fasting blood glucose of DO group rats decreased to (5.97±0.73)mmol/L from (23.04±3.51)mmol/L post-operative 8 weeks (P0.05) and keep the low level for weeks; the fasting GLP-1 of DO group raised to (23.82±2.07)pmol/L from (6.84±0.84)pmol/L post-operative 8 weeks (P0.05), much more significant at the post oral glucose 30 min condition. Conclusion The GBP operation has a significant treatment effect on type 2 diabetic rats, it plays an important role in the high secretion of GLP-1, which is caused by food into the lower segment of jejunum directly post-operatively.
