Observation on serum IL-17 family level variation of rabbit treated with allogeneic skin grafting after injection of recombinational human IL-10

Abstract

Objective This study is in order to observe serum IL-17 family level variation of rabbits treated with allogeneic skin grafting after injection of recombinational human IL-10, and also in order to investigate immunoreaction inhibition mechanism of recombinational human IL-10. Methods 30 healthy new zealand white rabbits were randomly and averagely distributed into 6 groups, which included group A(recombinational human IL-10 low-dose group, 5μg /(kg.d)), group B(recombinational human IL-10 high-dose group, 10μg /(kg.d)), group C(cyclosporine low-dose group, 5mg /(kg.d)), group D(cyclosporine highdose group, 10 mg /(kg.d)), group E(recombinational human IL-10 low-dose group, 5μg /(kg.d) cyclosporine low-dose group, 5mg /(kg.d)) and group F(normal saline blank control group, 0.9% sodium chloride solution 1ml/d), each group had 5 rabbits. These rabbits were continuously treated with drugs by intramuscular injection for 10 days from 1 d before skin grafting operation,middle ear arterial blood was respectively collected for rabbits of all groups on 1d before operation, 1d, 4d, 1 week, 2 weeks,3weeks and 4 weeks after operation. Serum was separated. Enzyme linked immunosorbent assay was used for detection concentration of IL-17 family members(IL-17A-F). Results Concentration of IL-17 A,IL-27(IL-17B/C/D),IL-17E(IL-25),IL-17 F for rabbits after allogeneic skin grafting operation was higher than that before operation, concentration of IL-17 A,IL-27(IL-17B/C/D),IL-17E(IL-25),IL-17 F reached to the peak value on 2 weeks after operation, then concentration decreased. rhIL-10 could inhibit secretion of IL-17 A,IL-27 and IL-17 F. inhibiting effect was strengthened with the increase of rhIL-10 dose, rhIL-10 could promote secretion of IL-25,no obvious dose-effect relationship was found. Conclusion Serum IL-17 family level of rabbit treated with allogeneic skin grafting after injection of recombinational human IL-10 was obviously improved,inhibiting effect of rhIL-10 on immunoreaction might be related to IL-17 family mediated inflammatory reaction and secretion of inflammatory factors.
