Study on cardiac protective effect of herbal compound Kuanxin on heart acute ischemia-reperfusion in rats

Abstract

Objective To investigate the cardiac protective effect of herbal compound kuanxin on heart acute ischemia-reperfusion in rats. Methods Thirty-two male healthful SD rats were divided into four groups randomly. Each group included eight rats,namely:sham operation control group (group C),ischemia-reperfusion model group (group M),herbal compound kuanxin group (group K) and trimetazidine group (group T). Ischemia-reperfusion was induced in rats by ligated the anterior descending branch of left coronary artery for sixty minutes and loosed it subsequently for thirty minutes. In the group C,the artery was exposed after the chest had been opened,but not ligated. All the rats were kept on fed for four weeks after the operation. Each rat in group K was fed with herbal compound kuanxin 14 ml/(kg·d) in addition,while each rat in group T was fed with trimetazidine 20 mg/(kg·d). Serum T-SOD,serum CPK activity,serum MDA and serum FFA content were measured seven days after operation. At the end of the experiment all rats were killed and heart tissue were collected for being examined under light micro-scope and electron microscope. Results Twenty-four hours after operation,serum CPK activity,FFA and MDA in groups M were significantly increased in comparison with those in group C (P0.01,P0.05),while serum T-SOD activity was significantly decreased compared with that in group C (P0.05). CPK and MDA in group T and group K were lower,T-SOD were higher than those in group M (all P0.05). But there was no difference between group T and group K (P0.05). Compared with group M,Serum concentration of FFA in group T was significantly lower (P0.05). The outcome from the light microscope and electronic microscope indicated that group C was nearly normal while there were obvious damages in group M. The damages ingroup T and K were much more slighter than that of group M.Conclusion Herbal compound kuanxin can scavenge oxygen free radicals and im-prove the structure mitochondria in order to take car-diac protective action,but it can not optimize myocardial metabolism.
