Abstract

Objective To explore the effects of sodium fluoride exposure on proliferation of human osteosarcoma MG 63cells.Methods MG-63 cells were exposed to 0,10-2,10-1,1,10,102,5×102,103,5×103,104,2×104and 4×104μmol/L NaF solution for 48 h.MTT assay was used to measure proliferation of MG-63 cells,and flow cytometry was applied to test the cell cycles.Results At the concentration of 102μmol/L NaF,MG-63 cells proliferation became increased;At 5×103μmol/L NaF,cells proliferation rate reached the peak,but at 2 ×104μmol/L,cells proliferation became inhibited.The three concentrations would be taken as the low-dose,moderate-dose and high-dose in the following experiments.After treatment by sodium fluoride for 48 h,cells in high-dose group had the highest apoptosis rate and the lowest PI(P0.05),while cells in moderate-dose group had the lowest apoptosis rate and the highest PI(P0.05).In term of the low-dose and control group,there was no significant difference in those two parameters.Conclusion Sodium fluoride has a double action to osteoblast.NaF can promote proliferation at lower concentration and inhibit proliferation at higher concentration.The double action may be related to the multiple types of pathological manifestations of skeletal fluorosis.
