The Study of Different Agtigen Retrieval Methods and Detectives of Detecting Expressing of CD117 in Gastrointestinal Stromal Tumor

Objective To study an ideal method of detecting expression of CD117 in gastrointestinal stromal tumors by immunohistochemistry. Methods CD117 antigen retrieval with four kinds of methods:① No antigen retrieval,②Citric acid buffer (PH6.0) microwave antigen retrieval,③EDTA buffer(PH8.0) microwave antigen retrieval,④0.1% trypsin digestion. CD117 antigen of tissue processed were detected by SP and PV staining. Results Positive cells of CD117 antigen using citric acid buffer (PH6.0) microwave antigen retrieval were higher than the other means and positive cells processed by PV staining were higher than SP staining. Conclusions it is the ideal method of detection of CD117 antigen that CD117 antigen was retrieved by citric acid buffer(PH6.0) microwave antigen retrieval and detected by PV staining.
