Establishment of Liver Transplantation Model in Rats by Two-Cuff Technique

Abstract

Objective To discuss the detail and points of rat orthotopic liver transplantation in experiments so as to establish a more efficient and stable animal model.Methods Based on the original two-cuff technique,we integrated varieties of modified techniques of rat orthotopic liver transplantation model,perfusion by abdominal aortic,evertedly suturing anastomosis suprahepatic inferior vena cava by single thread,anastomosis portal vein and infrahepatic inferior vena cava by cuff methold,end-to-end anastomosis common bile duct by single-tube intra-stent.we improved the details in surgery to reduce blooding,reasonably rewarm and rehydrate,shorten operative time and anhepatic phase.We observed postoperative survival rate and survival period.Results We established 120 cases(not including pretest)SD rat orthotopic liver transplantation model with improved two-cuff technique,the time of donor surgery was(17± 1.5) minutes,(37±2.5) minutes for receptor,the anhepatic phase was only(19±1.5) minutes,the success rate of surgery of liver transplantation reached 94.2% and the one week survival rate was 90%.Conclusion Skilled surgical operations,streamline operations,and improving the details,may improve the survival rate and survival period of rats,and establish a more efficient and stable model of rat orthotopic liver transplantation for study.
