Abstract
To find out the transmission route of the white spot syndrome virus(WSSV)during the nursing period of the tiger prawn,Penaeus monodon, we conducted a combined study using currently available nucleic acid diagnostic tools, including polymerase chain reaction (PCR) and in situ hybridization, and transmission electron (TEM) microscopy to examine the sites for virus multiplication. Upon condition that other transmission source of WSSV was removed in our experiment system except brooder with WSSV, the fertilized eggs from the infected brooder could fulfil the embryo development, metamorphosed into nauplius, zoea, mysis and post-larvae. The 2-steps WSSV PCR detection showed that the effected fertilized eggs spawned from the infected brooder were WSSV positive, and some of nauplius, mysis and post-larvae from the effected fertilized eggs were also WSSV positive. The WSSVmolecular probe detection (in situ) showed that the gills of the post-larvae from the effected fertilized eggs were WSSV positive, and the result was reconfirmed by TEM. These results represent that WSSV can be transmitted from the brooders to their post-larvae during the nursing period of the tiger prawn. At the same time, the results of the PCR detection on control were WSSV negative.
