Spectrophotometric Method for the Determination of Proteins with TZACA

Abstract

2-(2,3,5-triazolylazo)-1,8 dihydroxynaphthale-3,6-disulfonic acid reacts with proteins to form complexes in ClCH_2COOH-CH_3COONa buffer with pH 2.96. The maximum absorption wavelength was at 505.80 nm. The optimum reaction condition was investigated and a new method for the determination of protein was developed based on this binding reaction. Beer's law was obeyed in the range from 0 to 80 mg·L-1. The proposed method , which is rapid ,stable and of good simple selectivity and high sensitivity, has been applied to the spectrophotometric determination of total proteins in human serum samples with satisfactory results.
