Mechanism and Cytotoxic Effect of Maoeryan Polysaccharide on SMMC-7721 Strain Cells

Abstract

The human SMMC-7721 strain cells were given with maoeryan polysaccharide in vitro at the concentrations of 10.0,1.0 and 0.1 μg/mL for 48 h,respectively.The cytotoxic effect,the inhibitory rate for proliferation of SMMC-7721 strain cells and the apoptosis rate were determined by using flow cytometry,methods of MTT [3-(4,5-dimethyl thiazol-2yl)-2,5-diphenyltetrazolium bromide] and trypan-blue.The results showed that the maoeryan polysaccharide had significant cytotoxic effects on human SMMC-7721 strain cells.The inhibitory rates were 72%,47%,31%,and the apoptosis rate 26.4%,18.2%,16.3%,respectively,as compared with those of the control group.The maoeryan polysaccharide has significant cytotoxic effects on human SMMC-7721strain cells and their mechanism may be related to the inhibitation of cell division,and to the destruction of cell membrane and mitochondria.
