Abstract
The regenerative systems of alfalfa varieties(Millennium and Gannon No.4)were comparatively studied by using MS as basic medium adding different concentration of 2,4-D,KT and NAA.The results showed that the hypocotyl was the optimal explant material;2,4-D level had the less effect to callus induction of Gannon No.4,but evidently enhanced Millennium with the increasing concentration of 2,4-D.The effective media for inducing callus of these two variety was MS containing 2 mg/L 2, 4-D ,0.3 mg/L KT,30 g/L sucrose and 7.5g/L agar. The optimal differentiation medium was MS containing 0.5 mg/L KT,0.1 mg/L NAA and 20 g/L sucrose. The effective root inducing medium was 1/2 MS containing NAA0.5 mg/L.
