TLR4 is Involved in the Apoptosis of Hippocampus Cells in Cerebral Ischemia-reperfusion Mice

Abstract

Objective To study the effect of Toll-like receptor 4(TLR4) on the apoptosis in cerebral ischemia- reperfusion(I/R) mice. Methods 90 kun-ming mice were randomly divided into sham group, ischemia-reperfusion group and TLR4-blocked group in different time points of reperfusion (6h,12h, 24h, 48h and 72h). The expression of caspase-3 was studied by immunohistochemistry and Western blot, the apoptotic hippocampus cells were stained by TUNEL. Results Both the expression of caspase-3 and the number of apoptosis cells increased significantly in I/R group than in sham group(p 0.01), however the expression of caspase-3 and the number of apoptosis cells decreased in TLR4-blocked group compared with I/R group (p 0.01). Conclusion Blocking TLR-4 can decrease the cells apoptosis caused by ischemia-reperfusion.
