Study on the Determination of Quercetin and Kaempferol in Stamen Neulumbinis by High Performance Liquid Chromatography

Abstract

A high performance liquid chromatography(HPLC) method for the determination of quercetin and kaempferol in Stamen Neulumbinis was studied.The quercetin and kaempferol were extracted from the sample with 90% of methanol by heating circumfluence for 45 min.Then the extract was purified by solid phase extraction with Sep-Pak-C_(18) cartridge.The quercetin and kaempferol were separated on a ZORBAX Stable Bound(4.6 mm×(50 mm),3.5 μm) chromatographic column,with methanol and 0.2% phosphoric acid(50/50) as mobile phase at a flow-rate of 2.0 mL/min,and monitored with the photodiode array detector at 360 nm.The recoveries of the quercetin and kaempferol are 98%~102%,and relative standard deviations are 0.87%~1.22%.This method was applied to the determination of quercetin and kaempferol in Stamen Neulumbinis with good results.
