Abstract

OBJECTIVE To establish and study Fluorescence PCR method for clinical detection of Staphylococcus aureus(SAU) and methicillin-resistant Staphylococcus aureus(MRSAs).METHODS The drug susceptibility and specificity was performed by using standard fluorescence PCR method and procedure.RESULTS The highest value of the analytic sensitivity was 1×102 copies/reaction,meeting the requirements of the design, and the method was highly reproducible;the clinical sensitivity and specificity of detection of S.aureus from 1196 specimens by fluorescence PCR were 97.41% and 98.47%,respectively;the clinical sensitivity of detection of MASAU from 527 S.aureus samples was 99.74%,and the specificity was 95.59%;the detection rate of S.aureus from 385 nasal swab samples was 18.18%,and the detection rate of MRSA was 42.50% in these 40 S.aureus samples.CONCLUSION Fluorescence PCR has great advantages in clinical detection of S.aureus and MRSA.
