Abstract
Objective To determine the expression of LRP gene in acute leukemia patients and evaluate the relationship between LRP expression and multidrug resistance and prognosis.Methods Expression of LRP mRNA were determined in bone marrow cells from 47 patients and 7 normal subjects by semi quantitative reverse transcription polymerase chain reaction(RT PCR).Results Expression of LRP was negative in 7 normal subjects and was positive in 17 of 25 patients with acute myelocytic leukemia, in 1 of 12 patients with acute lymphocytic leukemia and 8 of 10 refractory or relapsed patients.An analysis demonstrated LRPmRNA level was significantly higher in newly diagnosed acute myeloid leukemia and refractory or relapsed patients than in normal subjects( P = 0.001 ),increased LRPmRNA level had correlation with lower sensitivity to initial chemotherapy with reduced overall survival rate ( P 0.05).Complete remission(CR)rate was lower in LRP positive patients(3/11)than in negative patients(4/5). Conclusion The expression of LRPmRNA could predict the treatment outcome and prognosis in acute myeloid leukemia. LRP was an important resistance factor and its determination could help us design chemotherapy project.
