Abstract
Objective To observe the effect of valsartan and spironolactone on the expression of hepatocyte growth factors in myocardium and vessel in Goldblott hypertensive rats.Methods Twenty four 6 week SD rats were divided into 4 groups at random:sham operated group,operated group,valsartan group and spironolactone group.Valsartan 30 mg·kg -1 ·d -1 ,spironolactone 20 mg·kg -1 ·d -1 were administered to rats in valsartan group and spironolactone group respectively.The expression of hepatocyte growth factor (HGF) mRNA in myocardium and vessel was determined by RT PCR.Results Levels of HGF mRNA/β actin mRNA in myocardium in valsartan group (1.17±0.08) and spironolactone group (0.85±0.08) were significantly higher than those in operated group (0.57±0.07) ( P 0.01,respectively),but lower than those in sham operated group (1.35±0.09).Levels of HGF mRNA expression in mesenterical arteries of two treatment groups were higher than those in operated group ( P 0.01),but lower than those in sham operated group ( P 0.01).Conclusion AngⅡ AT1 receptor antagonist and mineralocorticoid receptor antagonist prevented myocardial and vascular remodeling,accompanied by a significant increase in myocardial and vascular HGF.Overall,the increase in local HGF expression may play an important role in protection of target organs in Goldblott hypertensive rats.
