Abstract
To establish an improved and simplified hepatocyte perfusion technique for isolation of rat hepatocytes,we change the perfusion with collagenase in situ,adjust the perfusate and the perfusion condition,and apply the gradient centrifugation method to purify the hepatocytes.The results showed that rat hepatocytes isolated have high production,purity and viabilities.This new method is a effective and practical technique for isolating rat hepatocytes.
