Abstract

OBJECTIVE To study the effect of Sidiming(SDM)on glutamic acid(Glu)content of the brain cortex and hippocampus in morphine withdrawal rats induced by naloxone.METHODS A morphine dependent model of rats was established by subcutaneous injection(sc)of morphine in gradually increasing dosages. The brain cortex and hippocampus were collected from the morphine withdrawal rats induced by naloxone and the Glu content in the cortex and hippocampus was determined by HPLC with pre-column 2,4-dinitro-fluorobenzene(DNFB)derivatization.RESULTS Compared with the NS control group,Glu content in the cortex and hippocampus was not significantly changed in SDM control group, but it was significantly higher in the model group.Compared with the model group,Glu content in the cortex and hippocampus was significantly lower in both the SDM treatment group and the positive control clonidine group.CONCLUSION SDM can adjust the content of Glu in the cortex and hippocampus in the morphine withdrawal rats, which may be related to attenuating the morphine withdrawal symptoms.
