Abstract

Objective:This study was to investigate the effect of survivin on apoptosis and proliferation of human lung cancer line.Method:Survivin siRNA was transfected into lung cancer cells to inhibit the expression of survivin.mRNA and protein levels of survivin were detected by reverse transcription-polymerase chain reaction(RT-PCR) and Western Blotting.Proliferation inhibition rate of lung cancer cells was analyzed by colon forming assay.Cell apoptosis was assayed by flow cytometry(FCM).Result:(1)Survivin was ex-pressed in lung caner cells,but transfection of survivin siRNA could decrease the survivin expression in RNA and protein level;(2)Sur-vivin siRNA transfection could increase the rate of cell apoptosis;(3)Survivin siRNA could inhibit cell proliferation.Conclusion:Block-ade of survivin expression could enhance radiosensitivity through increasing rate of apoptosis and decreasing the proliferation in lung cancer cell.
