Abstract
To isolate the functional genes of specific expression in liver tissue of goose,the liver tissue was dissected from Jilin White goose,a Chinese indigenous breed,at 38 weeks of age.We used the SMART techniques to construct a full-length cDNA library with the long-distance PCR method to amplify double-strand cDNA.The modified vector of pBluescript Ⅱ SK plasmid with the adaptors containing SfiⅠsites was employed to recombine with the cDNA products amplified,and the recombinants were cloned by transformation into competent Escherichia coli DH2α.A plasmid cDNA library with goose liver tissue was constructed.The result showed that the titer of the goose liver tissue cDNA library is 1 123 pfu/μL,content of the library reaches 1.12×106 in which the percentage of recombinant clones is 96%.24 randomly selected clones were sequenced.The ratio of unigene obtained is up to 58.3%,and the percentage of complete cDNA sequences is 95.2%.It has laid the foundation for further study on the relevant gene structure,molecular biological function and expression regulation in goose.
