Abstract
A series of Escherichia coli strain K12 competent cells were produced in different conditions, including prepared methods, growth status, hot shocked time, etc.The effects of different factors on the efficiency of transforming plasmid pKD46 were studied.The highest transformation ability was 5.4×106 when the activated cells was cultured to an OD600 0.43, prepared with RbCl and hot shocked at 42℃ for 90 s.Meanwhile, the other factors affected the transformation efficiency were optimized.
