Abstract
Objective To investigate the transformation efficiency and affecting factors in the preparation of competent E.coli cells by using rubidium chloride (RbCl) method.Methods Competent cells prepared by RbCl were used to transform different types of plasmid. The effects of different strains of E.coli and different periods of freezing before transformation were analyzed, with the transformation efficiency of RbCl method compared with that of CaCl 2 method.Results Different types of plasmid were all efficiently tramsformed by RbCl method.For the same plasmid,different E.coli strains and different periods of freezing had no effect on transformation. The transformation efficiency of RbCl method was higher than that of CaCl 2 method.Conclusion The competent cells prepared by using RbCl method could be frozen for a long period without loss of transformation efficiency.
