Establishment of High Frequency Regeneration System of Stevia rebaudiana Bertoni Leaf

Abstract

Taking young leaves of Stevia rebaudiana Bertoni as explants,the effects of disinfection time and exogenous hormones on the survival rate,induction of in vitro shoot,growth and root formation were studied.The results showed that the optimum disinfection method was treated with 0.1%HgCl2 for 3 min;the optimum adventitious buds induction medium from leaves was MS+2.0 mg/L 6-BA+1.0 mg/L IAA+300 mg/L CH(Casein Acid Hydrolysate) and the average induction rate was 80%;the optimum shoot growth medium was MS+0.5 mg/L 6-BA+0.05 mg/L NAA+0.2 mg/L GA3+4.0 mg/L KT;the optimum rooting medium was 1/2MS+0.2 mg/L IAA with rooting rate 100%.The tissue cultured plants obtained grew well and had high survival rate after transplanting,the optimum regeneration system of Stevia rebaudiana Bertoni was found finally.
