Studies on the Rooting Culture in Plantlets of Stevia rebaudiana Bertoni Stem Apex and Transplanting

Abstract

The research was carried out using the tender stem apex of Stevia rebaudiana Bertoni as explants and MS medium as the basic medium.Poly shoots can be induced from the explants that were cultured on the basic medium supplemented with 6-BA 1.0 mg/L+Sucrose 30 g/L+Agar 6g/L,which gave a regeneration frequency of 100% if were not infected with germs.The developed medium was MS+6-BA 0.5 mg/L+NAA 0.05 mg/L+ Sucrose 30 g/L+Agar 6 g/L.We can obtain nearly 14-15 times poly shoots after it was cuhured in the developed medium 30 days.The best rooting medium is 1/4MS+IBA 0.1 mg/L+NAA 0.1 mg/L+Sucrose 30 g/L+Agar 6 g/L +active carbon 1 g/L,the rootage rate reached up to 100%.The test-tube plantlets needed to be trained two days by opening the lid.After transplanting the survival percentage was up to 95%.
