Abstract
Objective:To observe the effect of Chinese Herb recipe on the expression of ERK1/2,TGF-β1 gene,PDGF-B gene in early diabetic nephropathy rats,try to explore the possible mechanism of ERK1/2 activity and the protection role of Chinese Herb recipe on diabetic nephropathy.Methods:The diabetic nephropathy model of the rats were established by intra-abdominal injection of STZ.The experimental rats were randomly divided into normal contrast group,pattern group and treated group.Then the rats were treated with Chinese Herb recipe for six weeks.Blood glucose and kidney hypertrophy index were measured.The level of phospho-ERK1/2 was examined by western blot.TGF-β1 gene and PDGF-B gene were measured by RT-PCR.Results:Blood glucose,Kidney hypertrophy index and Extracelluar matrix of modern group and treated group were significantly higher than blood glucose,kidney hypertrophy index and extracelluar matrix of normal group after 6 weeks(P0.01).Treated group were lower than those of modern group(P0.01).The expression of pERK1/2,gene of TGF-β1 and PDGF-B of renal cortex in treated group were significantly lower than that of in modern group.Chinese Herb recipe can relieve the pathological change of renal tissue.Conclusions:①pERK1/2 was participated in the occurring and developing of diabetic nephropathy.②Chinese Herb recipe can down-regulate hyper-expression of gene of TGF-β1 and PDGF-B in renal cortex.③It may have relation with inhibiting the activity of pERK1/2 that Chinese Herb recipe can treat early diabetic nephropathy.④The activity of pERK1/2 may have relation with the hyperexpression of gene of TGF-β1 and PDGF-B of renal cortex.
