Effect of ATRA on proliferation and apoptosis of human BGC-823 gastric cancer cell

Abstract

Objective To observe the effect of all-trans retinoic acid(ATRA)on proliferation and apoptosis of human gastric cancer cell line BGC-823 and its mechanisms.Methods BGC-823 cells were treated with different concentration of ATRA.The cell proliferatiwas measured by methabenzthiazuron(MTT)assay and the apoptosis induced by ATRA was evaluated by TUNEL assay.The Survivin mRNA expression levels were detected by RT-PCR.Results ATRA inhibited growth of BGC-823 cells in a dose-dependent and time-dependent manner(P0.05).Cell apoptosis index increased gradually after exposure to ATRA for 24 h,48 h,72 h in a dose dependent manner(1 μmol/L)(P0.05).Survivin mRNA levels of BGC-823 cells treated with 1 μmol/L of ATRA were reduced over time.Conclusion ATRA inhibits the proliferation and induces apoptosis in human gastric cancer cell line BGC-823.The effect mechanism is related to arrest the expression of Survivin gene.
