Abstract
Castanea henryi was taken as material cultured in vitro to induce the plant regeneration.The experiments were conducted in order to select the appropriate explants for tissue culture and the method of optimal sterilization of explants.The results showed that the embryo was the optimal explant.The most optimal method of sterilization was dipping embryo in 70% alcohol for about 30 s and then in the solution of 0.2% HgCl_2 and tween-80 for 7-9 min;the most optimal method of inoculation methods was putting abaxial layer upwards.
