Abstract

Objective To study the inducement of apoptosis of prostate cancer cells by TNF related apoptosis inducing ligand (TRAIL).Methods Human prostate cancer cells were treated with 10 μg/L 100 μg/L TRAIL for 4 24 h.The apoptosis was detected by Annexin V fluorescence staining,TUNEL assay and transmission electron microscopy.The relationship between the percentage of apoptosis and the concentration of TRAIL was assayed by flow cytometry.The cellular proliferation inhibitory ratio was evaluated by MTT assay.Results Typical apoptosis morphological changes of the cells were observed after being treated by 10 μg/L 100 μg/L TRAIL for 4 24 h.With the increase of concentration of TRAIL or the prolongation of the treating time,the percentage of apoptosis was increased by 13.8% 79.6%. The proliferation inhibitory ratio of the cancer cells increased notably by 7.5% 37.9% when treated for 8 h, 16.7% 87.9% when treated for 12 h,and 39.7% 97.6% when treated for 24 h respectively.Conclusion TRAIL can effectively and quickly induce the apoptosis of prostate cancer cells.TRAIL may be a new therapy for advanced prostate cancer.
