Molecular Cloning and Sequence Analysis of Mulberry Phenylalanine Ammonia-lyase Gene

Abstract

Phenylalanine ammonia-lyase(PAL,EC 4.3.1.5) is the key and rate-limiting enzyme in plant phenylpropanoid pathway,and is considered to have a close relationship with plant resistance to diseases,biosynthesis of flavonoid pigments and other secondary substances.A cDNA fragment of pal gene was cloned successfully from mulberry(GenBank accession No.FJ938356).The cDNA was 1 105 bp long and encoded a polypeptide of 367 amino acids.Its 34th to 50th amino acids had the characteristic PAL domain.Homology analysis of mulberry pal gene with other plants revealed that it had the closest phylogenetic relationship with Prunus avium,with a sequence identity as high as 82%.Its sequence identity with Rubus idaeus was 80% and that with Populus trichocarpa and Coffea canephora was 75%,respectively.Therefore,the obtained mulberry cDNA sequence is one member of the pal gene family.
