Abstract
Background and purpose:Hypoxia is a common phenomena in tumors. The progression of tumor growth is associated with the expression of hypoxia-inducible factor-1α(HIF-1α) and vascular endothelial growth factor (VEGF). This study investigated the expression of HIF-α and VEGF in gastric carcinoma (GC) and studied the correlation between HIF-1α and VEGF in order to explore their roles in invasion and metastasis of gastric cancer. Methods:The expression of HIF-1 and VEGF were detected by streptavidin perotridase in 85 patients with gastric carcinoma. Results:⑴ The expression of HIF-1α in paired para-carcinoma was negative but in gastric carcinoma, the positive expression rate was 76.47% (65/85). The positive expression rates of HIF-1α in GC were significantly higher than that in paired para-carcinoma. ⑵ The expression of HIF-1α was significantly associated with the depth of invasion (P0.05),lymph node metastasis (P0.05),and TNM stage (P0.05), but it was not associated with the degree of differentiation (P0.05). ⑶The expression of VEGF in paired para-carcinoma was negative, but in gastric carcinoma, the positive expression rate was 60.00% (51/85). The positive expression rates of VEGF in GC were significantly higher than that in paired para-carcinoma. ⑷The expression of VEGF was significantly statistically associated with the depth of invasion (P0.05),lymph node metastasis (P0.05),and TNM stage (P0.05), but it was not associated with the degree of differentiation (P0.05). ⑸The positive rate of HIF-1α and VEGF was 76.47% and 60.00%, respectively,in which co-positive in 45 cases and the co-negative in 14 cases. The HIF-1α was correlated with the VEGF expression (r=0.367, P0.05). Conclusion:The expression of HIF-1α and VEGF were higher in the experimental group than in the paired para-carcinoma. Their expressions were correlated to the metastasis of gastric carcinoma and might play an important role in the gastric carcinoma. There had been a significant correlation between the positive expression rates of HIF-1α and VEGF. Therefore, the overexpression of HIF-1α induces tumor angiongenesis and invasion by activating the transcription of the VEGF gene. Therefore, the detection of the two proteins could be used as biomarkers to predict metastasis and prognosis of these patients.
