Abstract

Objective To study the neurotoxicity of amyloid beta protein on cultured PC12 cells before it aggregated into fibrillary protein.Methods The dose manner neurotoxicity of Aβ on PC12 cells was detected by flow cytometry, and the type of PC12 cell death was investigated by TUNEL staining, eletron microscope and DNA electrophoresis.Results With the increasing concentration of Aβ25 35 ,the ratio of cell death increased. 24 hours after 10 μmol/L Aβ25 35 treatment, TUNEL staining was positive, DNA electrophoresis showed DNA ladder , and electron microscope showed the nuclear condensation and bubbling in the cytoplasma. Conclusions Aβ demonstrated obvious neurotoxicity on PC12 cells before it aggregated, and induced PC12 cells apoptosis.
