Abstract
Two flavones were isolated and purified from Sophora japonica by high-speed counter-current chromatography(HSCCC) for the first time.The separation of the two flavones was improved by using ionic liquid.A two-phase solvent system composed of n-hexane ∶ ethyl acetate ∶ ethanol ∶ water ∶ acetic acid(1∶ 1∶ 1∶ 1∶ 0.05,v/v) was used.The results indicated that 18.2 mg of rutin and 9.6 mg of quercetin with their purities over 97% were obtained from 50 mg crude extract of Sophora japonica.By adding ionic liquid,the peak retention time was shorten to 55 min from the previous 85 min,the peak resolution was increased from 0.9 to 1.8.Hence,the addition of ionic liquid into HSCCC system contributed to achieve complete separation of these two flavones.The results can be used as the basis for future research.
