OPN expressions in laryngeal squamous cell carcinoma and effects on infiltration and metastasis by shRNA silence in cell Hep-2

Abstract

OBJECTIVE: To study the OPN expressions and detect its clinicopathological value in the largngeal squamous cell carcinoma(LSCC) tissues.To further study the biological effect of OPN down-regulation on the proliferation and invasiveness of laryngeal cancer Hep-2 cells.METHODS: SP immunohistochemical method was used to study the OPN expression on the routinely paraffirr embedded sections of surgical operated specimens of 44 laryngeal squamous cell carcinomas and adjacent tissues.Clinicopathological features were analysized.Lentivirus vector with RNAi small gene sequence of OPN(LV-shOPN) was transfected into Hep-2 cells.OPN expression was detected by Western blotting assay,the proliferation of Hep-2 cells was examined by MTT,transwell assay was used to detect the invasion ability of Hep-2 cells.RESULTS: The positive rate of OPN protein in 44 cases LSCC and adjacent tissues was 75.0%(33 cases) and 13.6%(6 cases) respectively,and the difference of OPN was statistically significant(P=0.000).The OPN expression levels were significantly correlated with tumor stage,grade and the presence of lymph node,not correlated with age,gender and tumor primary site.In vitro constructed LV-shOPN effectively inhibited the expression of OPN in Hep-2 cell(P=0.117) and the proliferation and invasiveness of Hep-2 cells(P=0.026;P=0.135).CONCLUSIONS: OPN expression maybe participate in the development and metastasis of LSCC,and may be important biological markers in reflecting the prognosis of the LSCC.LV-shOPN can inhibit the proliferation and invasiveness of Hep-2 cells,which is probably expected to provide a theoretical basis for treatment in largngeal carcinoma.
