Abstract
[Objective]It was necessary for Nesterenkonia sp. DF-1 to optimize the fermentation conditions for production of amylase in order to provide theoretical basis for the mass production of amylase in fermentation industry. [Method]The single factor method and multi-factor orthogonal experiment were adopted to investigate the effect of a set of variables,including fermentation time,temperature,initial pH,salinity,the change of carbon source and nitrogen source upon the influence of fermentation to produce amylase. [Result]When liquid medium volume was 100 ml / 250 ml,the optimal fermentation conditions were corn meal 1%,yeast extraction 2%,initial pH 8. 0,NaCl concentration 3%,temperature 37. 5 ℃ and shaking culture for 72 h at 160 r /min. [Conclusion]The best fermentation conditions for Nesterenkonia sp. DF-1 to produce amylase were obtained.
