Abstract

Objective To observe the application of self-made mouse monoclonal antibody(mAb) against human insulin-like growth factor-1(IGF-1) and discuss its application values in experimental research.Methods Purified mAb was used in Western blot test and immunohistochemical study of gastric cancer.Results Western blot analysis showed the mAb reacted specifically with IGF-1 which possessed biological activities and IGF-1 recombinant protein expressed by prokaryotic vector.Immunohistochemical experiments showed the positive rates of IGF-1 in gastric adenocarcinoma tissue were significantly higher than those in the normal gastric tissue.Conclusion The self-made monoclonal antibody would be useful for clinical application and basic research in IGF-1,and the overexpression of IGF-1 could be associated with the diagnosis of gastric adenocarcinoma.
