Abstract

Objective To investigate the effect of WY14643 on acute lung injury and its mechanism.Methods 128 male Wistar rats were randomly divided into four groups,including control group,acute lung injury group,LW 1 mg group and LW 3 mg group.Wistar rats were treated with lipopolysaccharide before intratracheal challenge with Wy14643.All the groups were killed at 1,2,4 and 8 h after LPS injection.TNF-α and IL-10 in lung were measured with ELISA.Results The level of TNF-α and IL-10 in supernatant liquid of rats lung homogenate and in hematoplasma at 2,4 and 8h in LW 1 mg group LW 3 mg group were significantly lower and higher than those in ALI group(P0.05).The level of MPO in ALI rats were significantly decreased.Conclusions The results suggest that Wy14643 may be useful to treat acute lung injury,whose mechanisms may involve that Wy14643 decreases the expression of TNF-α and increases the expression of IL-10.
