Abstract

Objective:To study the effect of different concentrations of agarose gel on recovery of digested plasmid vectors with different length. Methods:Two plasmids with different length were digested with two restriction enzymes and analyzed by electrophoresis of agarose gel with different concentrations. By comparing the relative position of plasmids before and after digestion,the effect of different concentrations of agarose gel on recovery of digested plasmid vectors was studied. Results:The relative position of plasmids before and after digestion changed in agarose gel with different concentrations,which were important for recovery of high purity of digested DNA fragments. The concentrations of agarose gel have different effects on plasmids with different length. Conclusion:Suitable concentration of agarose gel is important for recovery of digested plasmid vector.
