Effects of Shenmai Injection on Proliferation and Apoptosis of Lymphatic Endothelial Cells

Abstract

Objective To explore the influence of Shenmai Injection,a patent traditional Chinese medicine mainly composed of Ginseng,and Radix Ophiopogonis,on the proliferation and apoptosis of lymphatic endothelial cells(LECs).Methods LECs were isolated and cultured from healthy porcine thoracic duct,and they were identified with Ⅷ factor and vascular endothelial growth factor receptor-3(VEGFR-3).The inhibitory effect of Shenmai Injection on the growth of LECs was observed by methyl thiazolyl tetrazolium assay,and the apoptosis of LECs was detected by Hoechst33258 fluorescent staining.Results LECs were found to be typical.Shenmai Injection could significantly inhibit the proliferation of LECs within the concentration of 40 to 160 μg/ml,and the inhibitive effect showed a concentration-dependent manner.Hoechst 33258 fluorescent staining showed some apoptotic bodies around the nuclear of LECs treated with Shenmai injection.Conclusion Shenmai Injection can inhibit lymphangiogenesis,and induce the apoptosis of LECs.
