Abstract
AIM To investigate the effect of Zhouluotong Capsules(Astragali Radix,Cinnamomi Ramulus,Angelicae sinensis Radix,Rehmanniae Radix,Asari Herba,etc.) on TGF-β1/Smad3 signaling pathway in diabetic peripheral neuropathy mice.METHODS Forty KK/Upj-Ay mice were randomly divided into model,Zhouluotong Capsules(6.85,3.43,1.71 g/kg)groups.Ten C57BL/6 mice were selected as control group.Mice were given intragastrically for twelve weeks.Motor nerve conduction velocity was observed.The expressions of transforming growth factor-β1(TGF-β1),TGF-β1 receptor I(TGF-βRI),TGF-β1 receptor II(TGF-βRII),Smad3 were examined by real time PCR and Western blot.RESULTS Compared with the control group,motor nerve conduction velocity deceased and the expressions of TGF-β1,TGF-βRI,TGF-βRII,Smad3 increased in the model group(P0.01).Motor nerve conduction velocity increased and the expressions of TGF-β1,TGF-βRI,TGF-βRII,Smad3 significantly decreased in Zhouluotong Capsules group(P0.05,P0.01).CONCLUSION Zhouluotong Capsules have protective effects on diabetic peripheral neuropathy of mice via suppressing TGF-β1/Smad3 signaling pathway.
